I think this quote means that that feeling you get before you do something tells you if its right or wrong and that if you don't feel good while you're doing something then it's probably not the right thing to do. They can't make you change you life if you aren't willing to. For example if you have a drinking problem your family can tell you its bad for you and a million other things, but if you're not willing to change your life you won't. You have to be willing to change it no one can change for you.

You'll have a guilt conscience and you'll just keep on thinking about what you did wrong. If you spend a week studying for a big test and you ace the test your conscience will feel good and you'll be proud of yourself because you earned that grade with your hard work.

The first message tells you that you should never give up on your dreams just because you fail doesn't mean you have to give up. You have to try and try until you accomplish your dream/goal. Her second message tells you that it doesn't matter how old you are to be able to chase your dreams. Her third message tells you that without the help of others you are not very likely to succeed. Diana even though she failed to swim from Cuba to Florida 4 times before she didn't give up and even though she was 64 she chased her dream again and finally was able to accomplish it.
